My partner will be home today so we'll be able to get some good mid-cycle tests in a few hours. Is it 6 cycles that we should wait after the missed shot, to let the depot rebuild before we increase? I thought I read that somewhere but I could be wrong.

Yes, the depot can take 4-6 cycles to adjust/rebuild. See what he does today and you can take him up tomorrow, if necessary.
